Biochemical and molecular genetic analysis of SUPS5 TS mutant in yeast saccharomyces cerevisiae

TS mutants of SUPJ5 gene were selected through both in vivo and in vitro mutagenesis. The quantitative defect of the SUPS5 gene product measured by the suppression efficiency of the mutants using pinsmid borne lacZ gene fusion. The suppression of the mutants showed varry among the mutants and elevated by increasing the incubation temperature. The data suggested that mutation in each mutant occur on different site of the gene.
