Characterization and identification of thermostable alkaline lipase producing bacteria from hot spring around West Java

Eight isolates showed lipolytic activity on agar plate media containing olive oil and rhodamine B. Three best lipase producers, namely DMS3, KHA_P12, and KHA_T25, were further characterized and showed maximum specific activity at a mid exponential of growth phase. Identification of the isolates were performed through ribotyping method. Chromosomal DNA from these isolates were extracted and the 16S rRNA gene fragments were amplified, resulting fragment length at around 1.5 kb on agarose gel.
